ABSTRACT

OBJECTIVES@#To study the clinical characteristics of plastic bronchitis (PB) in children and investigate the the risk factors for recurrence of PB.@*METHODS@#This was a retrospective analysis of medical data of children with PB who were hospitalized in Children's Hospital of Chongqing Medical University from January 2012 to July 2022. The children were divided into a single occurrence of PB group and a recurrent PB group and the risk factors for recurrence of PB were analyzed.@*RESULTS@#A total of 107 children with PB were included, including 61 males (57.0%) and 46 females (43.0%), with a median age of 5.0 years, and 78 cases (72.9%) were over 3 years old. All the children had cough, 96 children (89.7%) had fever, with high fever in 90 children. Seventy-three children (68.2%) had shortness of breath, and 64 children (59.8%) had respiratory failure. Sixty-six children (61.7%) had atelectasis and 52 children (48.6%) had pleural effusion. Forty-seven children (43.9%) had Mycoplasma pneumoniae infection, 28 children (26.2%) had adenovirus infection, and 17 children (15.9%) had influenza virus infection. Seventy-one children (66.4%) had a single occurrence of PB, and 36 cases (33.6%) had recurrent occurrence of PB (≥2 times). Multivariate logistic regression analysis showed that involvement of ≥2 lung lobes (OR=3.376) under bronchoscopy, continued need for invasive ventilation after initial removal of plastic casts (OR=3.275), and concomitant multi-organ dysfunction outside the lungs (OR=2.906) were independent risk factors for recurrent occurrence of PB (P<0.05).@*CONCLUSIONS@#Children with pneumonia accompanied by persistent high fever, shortness of breath, respiratory failure, atelectasis or pleural effusion should be highly suspected with PB. Involvement of ≥2 lung lobes under bronchoscopy, continued need for invasive ventilation after initial removal of plastic casts, and concomitant multi-organ dysfunction outside the lungs may be risk factors for recurrent occurrence of PB.

ABSTRACT

OBJECTIVE@#To study the relationship between osteonecrosis of femoral head after internal fixation of femoral neck fracture and serum bone metabolism, vascular active factors, and analyze the risk factors.@*METHODS@#Total 150 patients with femoral neck fracture who underwent reduction and internal fixation from April 2016 to April 2019 were selected, including 83 males and 67 females. According to whether there was necrosis of femoral head after operation, they were divided into necrosis group(32 cases) and non necrosis group (118 cases). Before operation and 1, 3, 5 days after operation, the serum levels of beta-C terminal cross-linked telopeptides of typeⅠ collagen(β-CTX), N-telopeptide of typeⅠ procollagen(PINP), nitric oxide (NO), Endothelin-1 (ET-1) were measured. The clinical characteristics of the two groups were compared. The risk factors of postoperative femoral head necrosis were analyzed by logistic regression model. The value of serum indexes in predicting postoperative femoral head necrosis was analyzed by ROC curve.@*RESULTS@#There was no significant difference in the levels of serum PINP and β-CTX between necrotic group and non necrotic group before operation and 1, 3 and 5 days after operation(@*CONCLUSION@#The content of serum NO and ET-1 on the first day after operation can predict the necrosis of femoral head.

ABSTRACT

Nrf2 is the key transcription factor mainly for regulating oxidative homeostasis and cytoprotective responses against oxidative stress. Nrf2/Keap1 pathway is one of the most important cellular defense mechanisms against endogenous or exogenous oxidative stress. With its activation, a wide range of stress-related genes is transactivated to restore the cellular homeostasis. Recent studies revealed that the aberrant activation of Nrf2 is related to the malignant progression, chemotherapeutic drug resistance and poor prognosis. Nrf2 plays a crucial role in cancer malignancy and chemotherapeutic resistance by controlling the intracellular redox homeostasis through the activation of cytoprotective antioxidant genes. Nrf2 inhibitor containing many natural products has been deemed as a novel therapeutic strategy for human malignancies. This article reviews the progress of studies of the Nrf2/Keap1 pathway, and its biological impact in solid malignancies and molecular mechanisms for causing Nrf2 hyperactivation in cancer cells. In conclusion, we summarized the deve-lopment of Nrf2 inhibitors in recent years, in the expectation of providing reference for further drug development and clinical studies.

ABSTRACT

Inhibition of bitterness is a significant measure to improve the compliance and clinical efficacy of traditional Chinese medicine(TCM) decoction. According to the characteristics of TCM decoction, such as high dispersion of bitterness components, multi-component bitterness superposition and strong instantaneous stimulation, the research group put forward a new strategy to inhibit bitterness in the early stage based on the self-assembly characteristics of amphiphilic substances in aqueous solution, in order to reduce the distribution of bitterness components in real solution and achieve the purpose of bitter-masking. It was found that the bitter-masking effect of amphiphilic substances was different on the bitter compounds of various structures. Therefore, it was speculated that there might be a certain relationship between the bitter inhibition effect and the substrate structure. In this paper, the interaction between mPEG-PLLA and five bitter alkaloids(bamatine, jatrorrhizine, berberine, epiberberine and coptisine) in Coptidis Rhizoma was studied to explore the effect of substrate structure on the inhibition of bitterness. The sensory test of volunteers was used to determine the bitter-masking effect of mPEG-PLLA on the decoction of Coptidis Rhizoma and its main bitter alkaloids. The molecular docking and molecular force field were applied to locate the bitter groups and the bitter-masking parts. The relationship between the bitter strength and the structure was analyzed by the surface electrostatic potential of the bitter alkaloids, and the correlation between the bitter-masking effect and the structural parameters of the bitter components was explored by factor analysis, so as to clarify the structure-activity relationship of mPEG-PLLA in masking the bitterness of coptis alkaloids. It was found that mPEG-PLLA had significant taste masking effect on the decoction of Coptidis Rhizoma and five alkaloids. The masking effect was obviously related to the structure of different alkaloids: the effect increased with the increase of the number of hydrogen donors, rotatable bonds, molecular weight, and hydrophobicity, and decreased with the increase of surface electrostatic potential, electrophilicity and binding energy with bitter receptors. In this study, the influence of alkaloid structure of Coptidis Rhizoma on the butter-masking effect of mPEG-PLLA was preliminarily elucidated, providing a scientific basis for better exerting the bitter-masking effect of amphiphilic block copolymers.

ABSTRACT

OBJECTIVE@#To investigate the correlation between U2AF1 gene mutation and clinical manifestations and prognosis in patients with myelodysplastic syndromes (MDS).@*METHODS@#The clinical data of 203 MDS patients who accepted Next Generation Sequencing (NGS) was retrospectively analyzed in Nanfang Hospital, Southern Medical University from December 2012 to October 2019. According to whether the patients had U2AF1 gene mutation, the patients were divided into U2AF1 mutated group and non-mutated group, and the relationship between gene mutation characteristics and clinical manifestations and prognosis was analyzed. Then according to the difference of the mutation site of U2AF1, the patients in U2AF1 mutated group were divided into U2AF1@*RESULTS@#The incidence of U2AF1 mutation in MDS patients was approximately 11.3% (23/203), and the mutation frequency of U2AF1 allele was 32.5%. The male ratio in U2AF1 mutated group was significantly higher than that in U2AF1 non-mutated group (P=0.001). There was no patient who had complex karyotypes or TP53 gene mutation in U2AF1 mutated group. There were no significant differences in ages, blood parameters, bone marrow blasts, WHO 2016 classification, IPSS-R category, chromosomal abnormalities like del(5q), -7/del(7q), del(20q), +8, and gene mutation like ASXL1, DNMT3A, RUNX1, SF3B1, and SRSF2 mutation between U2AF1 mutated group and the non-mutated group. Compared with the non-mutated group, there was no significant difference in the overall survival time (P=0.377), the time of acute myeloid leukemia (AML) transformation (P=0.681), and the response rate to hypome- thylating agents in U2AF1 mutated group (P=0.556). Besides, no differences were observed in sex, diagnosis age, WHO 2016 classification, IPSS-R category, blood parameters, overall survival time, and AML transformation time between U2AF1@*CONCLUSION@#The U2AF1 gene mutation dose not affect the survival time, AML transformation time, and response rate to hypomethylating agents in MDS patients. Besides, there are no statistical differences in the clinical characteristics and prognosis of MDS patients between U2AF1

ABSTRACT

Three phenylpropanoid glycosides were purified by extensive chromatographic techniques including column chromatography over microporous resin, MCI, diol, sephadex LH-20, reverse phase C_(18) from water-extracts of Sophorae Tonkinensis Radix et Rhizoma. Their planar structures were elucidated by combination of various spectroscopic method, such as IR, UV, MS, and NMR. The absolute configuration of aglycone was determined by quantum chemical calculations. Their structures were elucidated as(8R)-3-(4-hydroxyphenyl)-1-propanol-2-O-β-D-glucopyranoside(1), kalopanaxin D(2),(E)-4-hydroxycinnamyl alcohol 4-O-[2'-O-β-D-apiofuranosyl(1″→2')]-β-D-glucopyranoside(3). Compound 1 was undescribed previously. Compounds 2 and 3 were firstly isolated from Sophora genus.

Respiratory infections,especially respiratory viral infections,are common diseases in children. Although most respiratory viral infection diseases are self-limiting,some children may be hospitalized or even life-threatened due to severe viral lower respiratory tract infection. With the progress of virology research,new and effective antiviral drugs have been developed. Thus,the study focuses on the therapeutic progress of respiratory viral infections so as to improve the treatment for common respiratory viral infections in children.

ABSTRACT

@#【Objective】To study the clinical characteristics,predisposing factors,prognosis and drug resistance of Staphylococcus aureus(SA)blood influenza infection in children,so as to provide basis for reasonable control and treatment of this disease.【Methods】182 cases of Staphylococcus aureus positive in blood culture from January 2016 to June 2018 were analyzed in terms of department distribution,infection type,basic diseases,clinical characteristics,antibiotic resistance,treatment outcome and prognostic factors. 【Results】 Staphylococcus aureus in blood culture was positive in 182 cases,mainly from neonatal SA(63 cases,34.6%)orthopaedics(22 cases,12.1%),PICU(20 cases,11.0%), hematological oncology(15 cases,8.3%),rheumatic immunology(15 cases,8.3%),and respiratory medicine(13 cases,7.1%);source of infection:community infection(109 cases,59.9%),hospital acquired infection(73 cases,40.1%);there were 103 cases with underlying diseases (56.6%);the most common initial symptoms were fever in 148 cases(81.8%),and methicillin-resistant staphylococcus aureus(MRSA)was found in 141 cases(77.5%);all staphylococcus aureus were resistant to penicillin ,ampicillin ,ampicillin/sulbactam ,ceftriaxone and clindamycin ,but no serious drug resistance was found. Prognosis:161 cases(88.5%)were cured and improved,21 cases(11.5%)were not cured and 21 cases(11.5%)died. The adverse prognostic factors were severe sepsis(χ2 = 35.613,P = 0.000)and mechanical ventilation(χ2 = 39.13,P = 0.000).【Conclusions】Patients with underlying diseases and newborns are the predisposing factors of blood stream infection of staphylococcus aureus,while those with severe sepsis and mechanical ventilation have poor prognosis. MRSA accounts for a high proportion of blood stream infection of staphylococcus aureus,and nosocomial infection is higher than community acquired infection. When empirical treatment of severe staphylococcus aureus bloodstream infection is ineffective,the treatment should be adjusted in time to take account of MRSA infection.

Objective@#To investigate the radiotherapy (RT)-induced changes in the brain structural network in patients with nasopharyngeal carcinoma (NPC).@*Methods@#Three-dimensional structural magnetic resonance data (3D-T1W) was adopted to investigate the structural network in 103 patients with NPC before and after receiving RT. The structural networks were then reconstructed using 3D-T1W. The radiation-induced changes in topology properties of small world network were analyzed by using graph theoretical analysis.@*Results@#Patients showed small world properties before and after RT. Compared with the pre-RT group, the global and local efficiency were lower, the shortest path length was longer and the clustering coefficient was less in the post-RT group. In addition, the hub regions in the post-RT group were significantly different from those in the pre-RT group, mainly located in the left rolandic operculum, right inferior frontal gyrus, right parahippocampal gyrus, right lingual gyrus, bilateral supramarginal gyrus, left superior temporal gyrus and temporal pole of the right middle temporal gyrus.@*Conclusion@#It is speculated that RT leads to high efficiency of network topology and information transmission, which provides a novel perspective for exploring the RT-induced brain changes, diagnosis of RT-induced injury and evaluation of RT efficacy.

ABSTRACT

OBJECTIVE@#To investigate the current status of empirical antibiotic therapy for children with Staphylococcus aureus sepsis and the effect of therapeutic paradigm on prognosis based on a retrospective analysis.@*METHODS@#A total of 78 children with Staphylococcus aureus sepsis who were admitted from January 2014 to August 2017 were enrolled. According to the preferred empirical antibiotics before the detection of Staphylococcus aureus by blood culture, these children were divided into a carbapenem group with 16 children, a β-lactam group with 37 children, a vancomycin group with 15 children and a vancomycin+β-lactam group with 10 children. A retrospective analysis was performed for related clinical data including general status, underlying diseases, Acute Physiology and Chronic Health Evaluation II (APACHE II) score, history of use of immunosuppressant, drug resistance to methicillin and prognosis. A logistic regression analysis was used to investigate the effect of empirical antibiotic therapy on the clinical outcome and prognosis of children with Staphylococcus aureus sepsis.@*RESULTS@#There were no significant differences among these groups in general status, underlying diseases, history of use of immunosuppressant, APACHE II score, nosocomial infection and detection rate of methicillin-resistant Staphylococcus aureus (P>0.05). There were significant differences in the incidence rate of septic shock and in-hospital mortality among these four groups (P<0.05). The carbapenem group had the highest incidence rate of septic shock and in-hospital mortality (69% and 50% respectively). The multivariate logistic regression analysis showed that empirical antibiotic therapy with different antibiotics had different risks for septic shock and in-hospital death in children with Staphylococcus aureus sepsis (P<0.05), and that an APACHE II score of ≥15 was an independent risk factor for septic shock in these children (P<0.05). The carbapenem group had significantly higher risks of septic shock and in-hospital death than the vancomycin group (P<0.05).@*CONCLUSIONS@#Inappropriate empirical use of antibiotics may lead to a poor prognosis in children with Staphylococcus aureus sepsis. Empirical use of carbapenems is not recommended for children suspected of Staphylococcus aureus sepsis.

ABSTRACT

<p><b>OBJECTIVE</b>To analyze the clinical manifestations, laboratory findings, treatment and prognosis of patients with hemophagocytic syndrome (HPS).</p><p><b>METHODS</b>A retrospective study was carried out to analyze the underlying disease, clinical characteristics, laboratory findings and outcomes of 46 patients with HPS.</p><p><b>RESULTS</b>This cohort included 19 cases of HPS secondary to cancer, 11 cases of HPS secondary to infection, 10 cases of suspected malignant lymphoma based on PET-CT findings (without biopsy), and 6 cases of unknown etiology. The coincidence rate of the clinical characteristics of the patients with the indices listed in HPS-2004 criteria were: fever (100%), elevated serum ferritin (100%), cytopenias (93.48%), splenomegaly (91.30%), hemophagocytosis in the bone marrow, spleen or lymph nodes (84.78%), hypofibrinogenemia (67.39%), and hypertriglyceridemia (54.05%). The cases of cancer, infections and unknown etiology showed significant differences in serum levels of ferritin and β2MG (P<0.05), and significant differences were found in triglycerides, LDH, and fibrinogenemia between the nonfatal and fatal cases (P<0.05).</p><p><b>CONCLUSION</b>HPS can be secondary to various underlying diseases, many associated with Epstein-Barr virus infection. Cancer, especially NK/T-cell lymphoma, is the main cause of HPS. Persistent fever, elevated serum ferritin level and cytopenias are the most sensitive indicators for diagnosis of HPS, and early diagnosis and treatment are critical to lower the mortality rate of this disease.</p>

ABSTRACT

OBJECTIVE: To investigate the role of cyclophilin A in the foaming process of macrophages induced by free silica( SiO_2). METHODS: The human peripheral blood mononuclear cell THP-1 in the logarithmic growth phase was induced and differentiated into human macrophages with phorbol 12-myristate 13-acetate at 100 μg/L for 48 hours. The cells were divided into 4 groups. The cells in the blank control group were not treated. The cells in the oxidized low-density lipoprotein( ox-LDL) control group were treated with a final concentration of 50 mg/L ox-LDL. The cells of 50 mg/L SiO_2 group were treated with 50 mg/L of SiO_2 and ox-LDL. The cells of 100 mg/L SiO_2 group were treated with 100 mg/L of SiO_2 and 50 mg/L of ox-LDL. After treatment of cells for 48 h,cell viability was measured by MTS method. The lipid accumulation of cells was observed by oil red O staining and colorimetric method; the expression of cyclophilin A in cell supernatant was detected by enzyme-linked immunosorbent assay,and the expression of cyclophilin A in the cells was detected by Western blotting. RESULTS: The cell viability was the highest when the concentration of SiO_2 was 100 mg/L compared with the control and other four SiO_2groups( P < 0. 01). The cell foaming change in the 100 mg/L SiO_2 group observed by oil red O staining significantly increased compared with the blank control group. The expression of total cholesterol,free cholesterol increased in the ox-LDL control group,50 mg/L SiO_2 group and 100 mg/L SiO_2group( P <0. 05),and the cholesterol specific gravity increased( P < 0. 05) compared with the blank control group,meanwhile the expression of cyclophilin A in the cell supernatant increased( P < 0. 05),and the expression in the macrophages cells decreased( P < 0. 05). CONCLUSION: Cyclophilin A is involved in the foaming process of macrophages induced by SiO_2.

ABSTRACT

Objective To analyze the health-related quality of life of elderly people living alone in Shanghai according to the survey data. Methods The data were extracted from the fifth national health services survey of Shanghai, and the data of 11 103 elderly people from 17 districts were analyzed. The elderly people living alone and those not living alone were matched using propensity score matching method, and the health-related quality of life of elderly people was measured by the European quality of life 5-dimensions (EQ-5D). The health-related quality-of-life indicators between the elderly people living alone and not living alone, including selection of EQ-5D dimensions, EQ-5D scores and EQ-VAS scores, were compared by Chi-square test and Mann-Whitney U test. Results The baselines of the elderly people living alone and not living alone matched well by propensity score matching, and there were no significant differences in selection of each dimension of the health-related quality of life, EQ-5D index scores or EQ-VAS scores between the two groups. Conclusion There is no difference in health-related quality of life between elderly people living alone and not living alone in Shanghai.

Objective To know about the two-week prevalence of the elderly people living in Shanghai and its influencing factors, so as to provide evidence for the health management of the elderly. Methods The data were extracted from the fifth national health services survey of Shanghai, and the data of 11 103 elderly people from 17 districts in Shanghai were investigated by multi-stage stratified cluster sampling, including the basic situation, healthy physical examination within a year and two-week prevalence. Univariate analysis was performed by Wilcoxon rank sum test and Kruskal-Wallis H test, and multivariate analysis was performed by logistic regression to explore the factors influencing the number of two-week prevalence of the elderly people aged 60 years and above in Shanghai. Results The total number of the two-week prevalence of the elderly people was 8 943, and the two-week prevalence rate was 805.458‰. The highest frequencies of two-week diseases were hypertensive and diabetes, accounting for 59.4% (5 314/8 943) and 14.9% (1 332/8 943), respectively. Results of univariate analysis and multivariate analysis showed that the main influencing factors of two-week prevalence were age, employment status, type of occupation, and physical examination within one year. Conclusion The two-week prevalence of elderly people in Shanghai is higher than the national average. Health guidance should be given to the factors influencing of the two-week prevalence in order to achieve a healthy aging.

@#AIM: To observe the clinical outcome of non-vascularization area after laser photocoagulation in the patients with retinopathy of prematurity(ROP)to lay the foundation for the clinical treatment of ROP. <p>METHODS: For a prospective follow-up observation from June 2014 to June 2016, 186 cases(372 eyes)underwent retinal laser photocoagulation were screened out in the ROP screening clinic and neonatal intensive care unit(NICU)bedside screening by the cooperative group of screening for ROP in our hospital. Non-vascularization area were exist in 26 cases(32 eyes). There were 17 male patients(18 eyes)and 9 females(14 eyes), the gestational age at birth was 29.4±0.4wk, and the average birth weight was 1222.8±70.3g. Among these cases, 10 patients(12 eyes)developed pre-threshold type 1 ROP, 12 patients(14 eyes)developed threshold ROP, and 4 cases(6 eyes)developed aggressive posterior-ROP. The Retcam fundus photography was performed at 1, 3, 8 and 12wk after laser surgery respectively. The changes of the non-vascularization area, the lesions and the additional lesions were observed. During following-up, if the lesions were found to increase, retinal laser photocoagulation or intravitreal injection of anti- vascular endothelial growth factor(VEGF)would be performed, until the lesions were completely under control. <p>RESULTS: In the all 26 patients, non-vascularization areas were found in 1wk after laser resection, but the lesions ridge dissipated from the stage 3 to stage 2 and stage 1. The dissipation were more obviously in patients with ROP of pre-threshold type 1. The range of non-vascularization areas was narrowed at 3wk in 27 eyes(84%), and the retinal vessels continued to develop to the surrounding areas,but in the other 5 eyes(16%),the non-vascularization areas were not significantly narrowed, the lesion ridge developed stage 3-4, with bleeding and proliferation phenomenon, then we timely to gave additional retinal laser photocoagulation. In these 5 eyes, 3(9%)developed threshold ROP, and 2 developed aggressive posterior-ROP. At the 8th week, 3 eyes(9%)of the 5 eyes were found with additional photocoagulation, the condition was controlled and stable. In 2 eyes(6%), the neovascularization disappeared in the ridge and the lesion was not found, then intravitreal injection of anti-VEGF were given. At the 12th week, the additional lesions in the 2 eyes had subsided, the ridge subsided, and the retinal blood vessels to the temporal shaped slightly stiff.<p>CONCLUSION: Retinal laser photocoagulation is an effective method for the treatment of retinopathy in premature infants. Most retinal blood vessels would develope in missing areas after the laser photocoagulation. A small proportion of patients need timely replenishment of laser photocoagulation treatment, if necessary, combined with intravitreal injection of anti-VEGF.

ABSTRACT

Objective To establish a method for the simultaneous determination of ursolic acid(UA)and oleanolic acid(OA) in Ziziphora clinopodioides Lam.,and the quantitative determination the UA and OA contents in the different Z. clinopodioides plant samples collected with various parts of the plant at different times,from different regions of Xinjiang,China. Methods Dual wave?length scanning method was used for the quantification of UA and OA spots on a silica gel G plate in the TLC analysis. The samples loaded on the silica gel G plate were in situ treated with the 1%iodine solution in dichloromethane,and then the plate was developed using cyclohexane,cyclohexane-chloroform-ethyl acetate-formic acid(20:5:8:0.1)as the developing solvent. In the dual wavelength scanning,the measurement wavelength was 530 nm and the reference wavelength was 700 nm. Results The UA and OA spots in samples were well separated on the TLC plate and could be simultaneously quantified by the present method. The average contents of UA and OA in Z. clinopodioides plant samples from 18 different areas were(1.84 ± 0.41)and(2.82 ± 0.89)mg/g,respectively. The contents of UA and OA in the plant increased from late spring to early summer and then decreased thereafter. As to the different parts of the plant,the contents of UA and OA were highest in leaves and lowest in stems. Conclusion The method is simple,fast and accu?rate. The present results provided basic data for further evaluation of the quality of Z. clinopodioides resources.

ABSTRACT

AIM:To investigate the effect of angiotensin II type 1 receptor (AT1R)-calcineurin (CaN) signa-ling pathway on the expression of sodium current channel Nav 1.5 at mRNA and protein levels in the hypertrophic ventricu-lar myocytes from neonatal rats .METHODS:The ventricular myocytes were isolated from the ventricles of 1-day-old neo-natal Sprague-Dawley rats and were divided into 4 groups according to different drug intervention as control group , pheny-lephrine (PE) group, losartan (Los)+PE group and cyclosporin A (CsA)+PE group.The method of RNA interference mediated by adenovirus carrying short hairpin RNA ( shRNA) was used to knock down the gene which encodes the beta subtype of CaN A subunit (CnAβ) and the cells were divided into 4 groups as Ad-Null group, Ad-Null+PE group, Ad-CnAβshRNA1 group and Ad-CnAβshRNA1+PE group.The mRNA expression of brain natriuretic peptide ( BNP) ,β-my-osin heavy chain (β-MHC) and Nav1.5 was detected by RT-qPCR.The protein levels of CnAβand Nav1.5 in the whole-cell extracts were determined by Western blot analysis .RESULTS:Treatment of the neonatal rat ventricular myocytes with PE for 24 h increased the protein-to-DNA ratio and the mRNA expression of BNP and β-MHC.The size of the cell surface was also increased after PE treatment .Treatment of the cells with PE increased the protein expression of CnAβ, and re-duced the protein expression of Nav 1.5.Both Los and CsA prevented those effects of PE .The mRNA expression of Nav1.5 was reduced by PE , and no significant difference of Nav 1.5 mRNA expression among PE group , Los+PE group and CsA+PE group was observed .Silencing of CnAβin the neonatal rat ventricular myocytes using Ad-CnAβshRNA1 inhibited the ability of PE to increase the mRNA expression of BNP , and diminished the ability of PE to reduce the protein expression of Nav1.5.CONCLUSION:AT1 R-CaN signaling pathway participates in regulating protein expression of Nav 1.5 in the hy-pertrophic ventricular myocytes from neonatal rats .

ABSTRACT

Objective:To investigate the effects of capecitabine and docetaxel on vascular endothelial growth factor and hepatocyte growth factor in patients with liver metastasis of breast cancer, and to analyze the possible mechanism of its treatment.Methods:A total of 216 patients with liver metastasis of breast cancer from January 2012 to June 2014 were enrolled,and the patients were randomly divided into observation group and control group with 108 cases in each group,and the control group was treated with docetaxel,and the observation group was treated with capecitabine combined with docetaxel.The short-term efficacy, vascular endothelial growth factor,hepatocyte growth factor,adverse reaction and survival rate were compared between the two groups.Results:The rate of disease control in the observation group was 88.89%,which was higher than that in the control group 70.37% (χ2=11.416,P<0.01). Vascular endothelial growth factor VEGF-A,VEGF-C,VEGFR-3 and hepatocyte growth factor (HGF) were significantly lower than those in the control group(t=14.684,16.726,16.733,4.508,P<0.05,P<0.01).There was no significant difference in adverse reactions between the two groups (20.37% vs 26.85%) (χ2=1.258,P>0.05).After the patients were followed up for 12 months,24 months,and 36 months,the survival rate of the observation group was higher than that of the control group,but the difference was not statistically significant (78.70% vs 70.37%,62.96% vs 55.56%,40.74% vs 30.56%) (χ2=1.976,1.227,2.442,P>0.05).Conclusion: Capecitabine combined with docetaxel treatment is helpful to improve the clinical efficacy,and safe and reliable.It may be related to the decrease of serum vascular endothelial growth factor and hepatocyte growth factor in patients.

ABSTRACT

Objective To research the effects of puerarin on Alzheimer′s disease(AD) rats spatial learning and memory dis‐order induced by Aβ1‐42 .Methods Bilateral hippocampal injection of Aβ1‐42 was used to induced AD model rats .All rats under‐went gavage administration with puerarin with different dose for 28 d since the 3rd day after the construction of model ;and the Morris water maze was used to test the spatial learning and memory ability of rats .Results The model group rats showed obvious learning and memory disorder ,and the ability of learning and memory disorder of rats in the high ,medium and low dose of puerarin interven‐tion group were significantly improved .Conclusion Puerarin can improve the spatial learning and memory ability of AD model rats .

ABSTRACT

<p><b>OBJECTIVE</b>To explore the inhibitory effect of estrogen against metastasis of human hepatocellular carcinoma MHCC97H cells and explore the molecular mechanism.</p><p><b>METHODS</b>The inhibitory effect of estrogen on the migration and invasion of MHCC97H cells was evaluated with wound healing assay and Transwell assay. Western blotting was used for investigating the expression of MMP-2, MMP-9, AKT and p-AKT in the cells treated with estrogen.</p><p><b>RESULTS</b>Estrogen treatment significantly inhibited the migration and invasion of MHCC97H cells in a dose-dependent manner. Estrogen significantly down-regulated the protein expressions of MMP-2 and MMP-9 and lowered the phosphorylation level of AKT.</p><p><b>CONCLUSION</b>The anti-metastatic effect of estrogen involves inhibition of MMP-2 and MMP-9 in MHCC97H cells probably by regulating AKT signal pathway.</p>
